Le Plerixafor (AMD3100) inhibe la chimiotaxie médiée par CXCL12 avec une puissance légèrement supérieure à son affinité pour CXCR4.

Il antagonise également la liaison du ligand SDF-1/CXCL12 avec une IC50 de 651 nM. Ce composé inhibe la liaison GTP médiée par SDF-1, le flux calcique médié par SDF-1 et la chimiotaxie stimulée par SDF-1 avec des IC50 de 27 nM, 572 nM et 51 nM, respectivement. Il n'inhibe pas le flux calcique contre les cellules exprimant CXCR3, CCR1, CCR2b, CCR4, CCR5 ou CCR7 lorsqu'elles sont stimulées par leurs ligands cognats, ni n'inhibe la liaison du récepteur de LTB4. En soi, il n'induit pas de flux calcique dans les cellules CCRF–CEM, qui expriment plusieurs GPCRs, y compris CXCR4, CCR4 et CCR7.

Pour les études de liaison compétitive contre CXCR4, le Plerixafor (AMD3100) est incubé à une gamme de concentrations pendant 3 heures à 4 °C dans un tampon de liaison (PBS contenant 5 mM MgCl2, 1 mM CaCl2, 0,25 % BSA, pH 7,4) avec 5 × 105 cellules CCRF–CEM et 100 pM de 125I-SDF-1α (2200 Ci/mmol) dans des plaques filtrantes Milipore DuraporeTM. Le 125I-SDF-1α non lié est éliminé par lavage avec 50 mM HEPES froid, 0,5 M NaCl pH 7,4. L'essai de liaison compétitive contre BLT1 est effectué sur des membranes de cellules CHO-S exprimant le BLT1 recombinant. Les membranes sont préparées par lyse cellulaire mécanique suivie d'une centrifugation à grande vitesse, resuspendues dans un tampon 50 mM HEPES, 5 mM MgCl2 et congelées rapidement. Ce composé est incubé avec la préparation membranaire pendant 1 heure à température ambiante dans un mélange de dosage contenant 50 mM Tris, pH 7,4, 10 mM MgCl2, 10 mM CaCl2, 4 nM LTB4 mélangé avec 1 nM 3H-LTB4 (195,0 Ci/mmol) et 8 μg de membrane. Le 3H-LTB4 non lié est séparé par filtration sur des plaques filtrantes Millipore Type GF-C.
In vivo

Chez les souris diabétiques, une seule application topique de Plerixafor (AMD3100) favorise la cicatrisation des plaies en augmentant la production de cytokines, en mobilisant les cellules EPC de la moelle osseuse et en améliorant l'activité des fibroblastes et des monocytes/macrophages, augmentant ainsi l'angiogenèse et la vasculogenèse.

Des cohortes de souris reçoivent du PBS, de l'IGF1, du PDGF, du SCF ou du VEGF pendant cinq jours consécutifs et ce composé le 5e jour. Le nombre et la taille des colonies sont les plus élevés chez les souris injectées avec de l'IGF1 plus ce composé par rapport aux groupes traités avec le PDGF, le SCF et le VEGF, en combinaison avec le Plerixafor.
