Description La Dihydromyricetin (Ampelopsine, Ampeloptine) est un flavonoïde antioxydant naturel extrait d'Ampelopsis grossedentata.
Cibles
GABAA receptor
In vitro La Dihydromyricetin présente une activité antioxydante en tant que piégeur efficace du radical 1,1-diphényl-2-picrylhydrazyle (DPPH). Elle inhibe également l'augmentation des valeurs de peroxydation lipidique (LPO) dans le système d'acide linoléique catalysé par FeSO4-acide édétique. Le DHM (1 μM), en tant que nouveau médicament anti-intoxication alcoolique, antagonise à la fois la potentialisation aiguë des GABA(A)Rs induite par l'EtOH et la plasticité des GABA(A)R induite par l'exposition/le sevrage à l'EtOH, et augmente l'expression de la sous-unité alpha4 du GABA(A)R dans l'hippocampe et les neurones cultivés.
In vivo La Dihydromyricetin (1 mg/kg, i.p.) contrecarre l'intoxication alcoolique aiguë (EtOH), ainsi que les signes de sevrage chez les rats, notamment la tolérance, l'anxiété accrue et la susceptibilité aux crises. LD50 : Souris 1.41g/kg (i.p.)
