Description

Cette lactone stéroïdienne, isolée de Withania somnifera, inhibe puissamment l'activation du NF-κB en empêchant l'activation, induite par le facteur de nécrose tumorale, de l'IκB kinase β via un mécanisme redox sensible à la thioalkylation. Elle se lie également à la protéine de filament intermédiaire (FI), la vimentine, avec une activité antitumorale et anti-angiogénique.

La Withaferine A (WFA) inhibe puissamment l'activation du NFκB en empêchant l'activation, induite par le facteur de nécrose tumorale, de l'IκB kinase β via un mécanisme redox sensible à la thioalkylation. Elle se lie à la protéine de filament intermédiaire (FI), la vimentine, en modifiant de manière covalente son résidu cystéine, qui est présent dans le domaine coiled-coil 2B α-hélicoïdal hautement conservé, et induit l'agrégation des filaments de vimentine in vitro.

In vivo

La Withaferine A (WFA) présente clairement des caractéristiques anti-inflammatoires dans un modèle murin d'inflammation aiguë. Son inhibition induite de la croissance capillaire dans un modèle murin de néovascularisation cornéenne est compromise chez les souris déficientes en vimentine.
