L'essai kinase Lck HTRF implique la phosphorylation ATP-dépendante d'un peptide substrat biotinylé de gastrine en présence ou en absence d'un composé inhibiteur. La concentration finale de gastrine est de 1,2 μM. La concentration finale d'ATP est de 0,5 μM (Km app = 0,6 ± 0,1 μM), et la concentration finale de Lck (une fusion de domaine GST-kinase (AA 225−509)) est de 250 pM. Les conditions de tampon sont les suivantes : 50 mM HEPES pH = 7,5, 50 mM NaCl, 20 mM MgCl2, 5 mM MnCl, 2 mM DTT, 0,05 % BSA. L'essai est éteint et arrêté avec 160 μL de réactif de détection. Les réactifs de détection sont les suivants : Tampon composé de 50 mM Tris, pH = 7,5, 100 mM NaCl, 3 mM EDTA, 0,05 % BSA, 0,1 % Tween20. Avant la lecture, de la streptavidine allophycocyanine (SA-APC) est ajoutée à une concentration finale dans l'essai de 0,0004 mg/mL, ainsi que de l'anticorps anti-phosphotyrosine eurolaté (Eu-anti-PY) à une concentration finale de 0,025 nM. La plaque d'essai est lue dans un lecteur de plaques à fluorescence Discovery avec une excitation à 320 nm et une émission à 615 et 655 nm. Les essais pour d'autres kinases sont réalisés de manière similaire à celle décrite ci-dessus, en faisant varier les concentrations d'enzyme, de substrat peptidique et d'ATP ajoutés à la réaction, en fonction de l'activité spécifique de la kinase et des Km mesurés pour les substrats.
