La Kd pour la liaison d'Onalespib (AT13387) est de 0,7 nM. Cela se compare à une Kd de 6,7 nM pour la liaison de l'ansamycine 17-AAG au même site. La stœchiométrie moyenne de liaison pour ce composé est de 1,03. Son inhibition d'un certain nombre de kinases isolées est également étudiée, y compris CDK1, CDK2, CDK4, FGFR3, PKB-b, JAK2, VEGFR2, PDGFRβ et Aurora B. Aucune des kinases testées n'est significativement inhibée à des concentrations inférieures à 30 μM. C'est un puissant inhibiteur de la prolifération et de la survie de nombreuses lignées cellulaires différentes (telles que la lignée cellulaire MES-SA) issues d'une variété de types de tumeurs différents. Sur un panel de 30 lignées cellulaires tumorales, il inhibe puissamment la prolifération cellulaire avec des valeurs de GI50 comprises entre 13 et 260 nM. Le composé inhibe également la prolifération de la lignée cellulaire épithéliale prostatique humaine non tumorigène PNT2 avec une valeur de GI50 de 480 nM.

Les valeurs de Kd pour la liaison d'Onalespib (AT13387) à HSP90 sont déterminées avec un format de calorimétrie isotherme (ITC) de compétition. Les expériences ITC sont réalisées sur un Micro Cal VP-ITC à 25 °C dans un tampon comprenant 25 mM Tris, 100 mM NaCl, 1 mM MgCl2 et 1 mM Tris(2-carboxy-éthyl)phosphine à pH 7,4 afin de maintenir l'affinité plus élevée.

Lorsqu'il est administré de manière intermittente, Onalespib (AT13387) peut être toléré à des doses allant jusqu'à 70 mg/kg deux fois par semaine ou 90 mg/kg une fois par semaine. La perte de poids corporel chez les souris ne dépasse pas 20 % avant de se rétablir dans tous les cas, sauf un, et la perte est la plus élevée après la deuxième dose. L'inhibition de la croissance tumorale est similaire chez NCI-H1975 pour les deux régimes posologiques. Le maintien des effets antitumoraux avec une période d'arrêt du traitement aussi prolongée est compatible avec l'action pharmacodynamique prolongée de ce composé observée pour l'EGFR mutant et d'autres biomarqueurs in vitro et in vivo, ainsi que la rétention prolongée de celui-ci dans les tumeurs.

How to prepare it for in vivo studies?

It can be dissolved in 2% DMSO/30% PEG 300/ddH2O at 10 mg/ml as a clear solution. But after stayed for about 1 hour, the precipitation will goes out. So it is recommended to be prepared before use.
