Docetaxel est un agent cytotoxique, en particulier pour les cellules proliférantes, ce qui est lié à sa capacité à promouvoir la formation de faisceaux de microtubules et à induire un arrêt mitotique soutenu, suivi de l'apoptose des cellules arrêtées en mitose ou d'un blocage mitotique permanent. Ce composé supprime l'instabilité dynamique des microtubules ainsi que le « tread-milling », entraînant l'incapacité des chromosomes à se séparer dans les cellules filles, ce qui déclenche à son tour une sortie prématurée de la mitose plutôt qu'un blocage à cette phase du cycle cellulaire. Il inhibe la survie clonogénique des cellules cancéreuses humaines Hs746T (estomac), AGS (estomac), HeLa (col de l'utérus), CaSki (col de l'utérus), BxPC3 (pancréas), Capan-1 (pancréas) avec une IC50 de 1 nM, 1 nM, 0,3 nM, 0,3 nM, 0,3 nM et 0,3 nM respectivement. Cet agent inhibe la migration des cellules endothéliales qui n'affecte pas la morphologie macroscopique des microtubules ni n'inhibe la prolifération cellulaire, bien qu'il produise des effets plus subtils sur la dynamique des microtubules. Il inhibe la migration des HUVEC avec une IC50 observée de 1 pM. La chimiotaxie des HUVEC stimulée par l'un des deux facteurs angiogéniques, la thymidine phosphorylase ou le VEGF, est inhibée par ce produit chimique avec une IC50 de 10 pM et est abolie à 1 nM. Il induit l'expression de la Prostaglandin H Synthase-2m (PGHS-2) dans les monocytes humains, mais pas dans les macrophages murins RAW 264.7.

Docetaxel (33 mg/kg/dose, administré par voie intraveineuse tous les 4 jours pendant 3 injections) entraîne un retard de croissance tumorale de 19,3 jours dans les xénogreffes de côlon M2OL2. Ce composé montre également de grandes activités antitumorales dans les xénogreffes MX-1, SK-MEL-2, LX-1 et OVCAR-3. Il inhibe la réponse angiogénique au facteur de croissance fibroblastique 2 avec une IC50 de 5,4 mg/kg lorsqu'il est injecté deux fois par semaine sur une période de 14 jours, et l'angiogenèse est complètement bloquée chez les souris qui reçoivent 10 mg/kg de ce produit chimique. Il présente une sélectivité pour la migration des cellules endothéliales et/ou la formation de microvaisseaux car l'infiltration des cellules inflammatoires dans le bouchon de Matrigel est beaucoup moins sensible à l'inhibition par ce composé.
